Insufficient leptin signaling to hypothalamus causes overfeeding which finally results in hyperglycemia followed by insulin resistance. Leptin resistance makes people obese. Adipose tissue secrets pro-inflammatory cytokines (interleukin-6, tumor necrosis factor-α) which can alter gene expression of signal transducer and activator of transcriptions, suppressor of cytokine signaling. Increased suppressor of cytokine signaling 3 inhibits the phosphorylation of insulin receptor substrate which finally leads to insulin resistance
